With regard to the mentioned peculiar toxicities, it is a known fact that majority of frontline chemo drugs alone are hard & toxic on the liver (Transamine, elevation of hepatic enzymes into bloodstream) Cytarabine is no exception as per: - https://www.ncbi.nlm.nih.gov/pubmed/2178634
Also mentioned is relatively frequent, but does not represent therapeutic problem.
Goodness me appendicitis...seriously...I've suffered that after a decent bout of the flu let alone full on chemotherapy ..
Which is why the team have chosen to slightly lower Cytarabine dose to find the optimum combination that suits a broader target.
I do not see this as a PTX200 toxicity issue, PTX200 has already proven to be low toxicity at current dosing levels, just a matter of finding that sweet spot in combination therapy.
I bet if you asked the 4 CR & many other PR & SD patients in PXT200 trials what they think of the drug? I'm sure they all would call it a miracle that has saved or bettered their quality of life, whereby nothing else on the market can achieve this result.
