Anything to do with the central nervous system has a higher risk than other biotech stuff. This is especially true of anything involving cognitive testing where variability is so high.
I was involved with a US biotech for a while which thought it had a great new drug for the treatment of stroke. It got all the way to phase III before failing to prove efficacy...
